Overview: Ipamorelin can release growth hormone, but wellness claims outrun the trials. Review human evidence, FDA concerns, Australian status and 2026 WADA rules.
On this page
Ipamorelin can stimulate growth-hormone release. That does not make it a proven anti-ageing or body-composition therapy.
Human studies establish short-term pharmacology and include one disease-specific postoperative-ileus programme. They do not establish the wellness, recovery, muscle-building or longevity claims commonly attached to online products.
| Evidence question | Best available answer | Confidence |
|---|---|---|
| Does it activate the ghrelin receptor? | Yes. Ipamorelin is a GHSR1a agonist and growth-hormone secretagogue. | Established pharmacology |
| Does it release GH in humans? | Yes, in controlled short-term pharmacology studies. | Human biomarker evidence |
| Does it treat postoperative ileus? | A phase 2 trial studied this specific question; it did not create a general approved use. | Disease-specific human trial |
| Does it improve recovery, muscle or longevity? | Not established by robust clinical outcome trials. | Unsupported extrapolation |
| Is long-term safety defined? | No. Regulators identify important data and product-characterisation gaps. | Major uncertainty |
What is ipamorelin?
Ipamorelin is a synthetic pentapeptide that activates the growth-hormone secretagogue receptor. This receptor is also activated by ghrelin. Experimental receptor activity can trigger pituitary growth-hormone release, but a hormone response is an intermediate biological measure rather than a patient-relevant outcome.
What has been studied in humans?
The healthy-volunteer pharmacokinetic–pharmacodynamic study examined ipamorelin exposure and growth-hormone responses. A hormone response demonstrates pharmacological activity; it does not establish a durable health benefit.
The later randomised postoperative-ileus trial studied gastrointestinal recovery after bowel resection. Its results must be interpreted in that population and setting.
| Feature | What the paper reports |
|---|---|
| Design | Multicentre, double-blind, placebo-controlled; intravenous ipamorelin after bowel resection |
| Participants | 117 enrolled; 114 in the safety and modified intention-to-treat populations |
| Key efficacy measure | Time to tolerance of a standardised solid meal |
| Median time | 25.3 hours with ipamorelin; 32.6 hours with placebo; p = 0.15 |
| Authors’ efficacy conclusion | No statistically significant difference in the key or secondary efficacy analyses |
The numerical difference is not proof of an effective treatment. A nonsignificant result also does not prove exact equivalence. This small, disease-specific study cannot establish body-composition, anti-ageing, sleep or athletic-recovery benefits, nor define long-term safety. Original study records checked: 22 October 2026.
What does “selective” really mean?
Ipamorelin is often described as selective because experimental studies reported a different hormone-release pattern from some older growth-hormone secretagogues. Selectivity does not mean side-effect-free. It describes activity observed under particular experimental conditions; it does not replace long-duration comparative safety data.
Three separate questions
Human pharmacology says yes.
Can a product be identified?
That requires validated analytical and manufacturing evidence.
That needs clinical outcomes in the intended population.
Is the benefit worth the harm?
That needs adequate duration and safety comparison.
FDA safety concerns
The FDA’s current compounding safety table says ipamorelin acetate may pose immunogenicity risks because of aggregation or peptide-related impurities. It also notes that unnatural amino acids complicate characterisation and that safety information is insufficient for several injectable routes. FDA advisory material discussed serious events in the postoperative-ileus development programme, including deaths whose causal relationship remained uncertain.
| Risk area | Why it matters | Evidence limit |
|---|---|---|
| Aggregation and impurities | May affect immunogenicity and exposure | A label or simple purity percentage is insufficient |
| Unnatural amino acids | Increase analytical-characterisation complexity | Identity must be demonstrated, not assumed |
| Route-specific safety | Risk can differ by formulation and administration route | Data cannot be transferred automatically |
| Long-term GH-axis exposure | Persistent hormonal effects require outcome and safety follow-up | Short pharmacology studies are inadequate |
Australian regulation and advertising
The October 2026 Poisons Standard names ipamorelin in Schedule 4 and Appendix D, clause 5, at item 25. Regulatory source checked: 22 October 2026. The TGA’s 2026 advertising guidance includes ipamorelin when explaining that prescription medicines cannot be advertised to the public through direct or substitute language. These rules do not establish an approved indication or validate an online product.
Sport status
The 2026 WADA Prohibited List names ipamorelin among growth-hormone secretagogues prohibited at all times. Athletes should use current official anti-doping resources for decisions affecting eligibility.
Frequently asked questions
Is ipamorelin a peptide?
Yes. It is a synthetic pentapeptide and a growth-hormone secretagogue.
Has ipamorelin been tested in humans?
Yes, including short-term pharmacology studies and a disease-specific postoperative-ileus trial. Those studies do not prove broad wellness or performance claims.
Does selectivity mean it has no cortisol or prolactin effects?
No universal conclusion follows from the word “selective.” Results depend on study design, population, exposure and what was measured.
Is ipamorelin approved for anti-ageing in Australia?
The reviewed official sources do not establish an ARTG-approved anti-ageing indication. Schedule 4 status and experimental study are not approval.
Is ipamorelin allowed in sport?
WADA names it among growth-hormone secretagogues prohibited at all times.
